Data Automation and Biotherapeutic Discovery

Generating biotherapeutics is a complex process requiring interrogation of thousands of drug candidates. As demand for complex formats and mechanisms increases, so does the demand for thorough validation of biologic molecules. Many attempts to automate this process have been undertaken with the adoption of liquid handlers, scheduling software, and fully independent production devices. Although much enthusiasm has been directed at reducing work at the bench, data analysis and presentation represent some of the most labor-intensive components that lead to experimental success. Boehringer Ingelheim Biotherapeutics Discovery has undertaken several data automation efforts to increase efficiency, reduce cost of experimentation, and improve decision making for biotherapeutic screening. Nanobody, or VHH, has gained a lot of interest for its strong epitope interaction, high stability, and tissue penetration capability. Here we will demonstrate how the Twin-Strep Tag technology and the Gator Pro system can efficiently express, purify, and screen a panel of nanobodies to select the binding candidate for CD45 antigen, a membrane protein.

Engineering PROteolysis-Targeting AntiBodies (PROTABs) for degradation of cell surface receptors

E3 ubiquitin ligases ubiquitinate and degrade target proteins. Here we describe Proteolysis Targeting Antibodies (PROTABs) which engage transmembrane E3 ubiquitin ligases and targets to achieve targeted protein degradation. We demonstrate tumor-selective activity with ZNRF3 and RNF43, two Wnt-dependent E3 ligases overexpressed in CRC. We further show the approach can be expanded to multiple targets and ligases, and through engineering antibody formats define the ground rules for antibody-based targeted protein degradation.
